A practical reference on Cake appearance: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Formulation composition influences whether freeze-drying produces an intact cake or a collapsed mass. Excipients such as sugars and polymers can raise the collapse temperature and provide bulk during drying. The critical temperature for primary drying is often the collapse temperature or the glass transition temperature of the maximally concentrated phase. If the product temperature exceeds this threshold, the frozen matrix may soften and lose structure. Established practice therefore links shelf temperature and chamber pressure to the formulation's thermal properties.
The physics of freeze-drying couples heat transfer, mass transfer, and phase change. Heat supplied through the shelf must reach the sublimation front without melting the ice or degrading the product. Water vapor then travels through the already dried layer and leaves the chamber, where low pressure and cold traps keep it from returning. The dried layer acts as a resistance to vapor flow, so drying rate changes as the front recedes. Open questions remain about how pore structure and formulation heterogeneity affect drying uniformity at larger scales.
Lyophilization removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts liquid water into ice and concentrates dissolved solids. Primary drying then lowers chamber pressure so ice changes directly into vapor without passing through a liquid phase. Secondary drying raises the shelf temperature to remove bound water that remains after ice sublimation. The result is a dry, porous structure that can be reconstituted later.
Analytical methods for lyophilized solids must account for the low moisture content and the fragile cake. Karl Fischer titration is widely used for water content, while near-infrared spectroscopy can measure moisture non-destructively in sealed containers. X-ray diffraction and modulated differential scanning calorimetry help identify crystalline or amorphous phases. Residual solvent analysis may be needed if organic solvents were used during formulation. The combination of these methods supports batch release and long-term stability assessment.
Lyophilized products are typically stored as sealed solids in vials or syringes. Moisture ingress is a major concern because many dried cakes are hygroscopic and can lose stability when exposed to humid air. Storage temperature depends on the formulation; some products are kept refrigerated, while others are stable at room temperature. Container closure integrity and headspace moisture are often monitored. Light protection may also be required for some photosensitive materials.
Quality control for lyophilized materials includes visual inspection, residual moisture measurement, and reconstitution testing. Cake appearance can reveal process problems such as collapse, shrinkage, or meltback, although appearance alone does not prove potency. Residual moisture is commonly measured by Karl Fischer titration or by loss on drying. Reconstitution time is checked because a slow or incomplete dissolve can indicate a change in pore structure. Stability studies track these attributes over time under defined temperature and humidity conditions.

=== Red blood cell transfusions === Blood transfusions are the main treatment approach for prolonging life. Donated healthy red blood cells have a functional life of 4 to 6 weeks before they wear out and are broken down in the spleen. Regular transfusions every three to four weeks are necessary in order to maintain hemoglobin at a healthy level. Transfusions come with risks including iron overload, the risk of acquiring infections, and the risk of immune reaction to the donated cells (alloimmunization).
Hundreds of additional cases of liver complications in people taking bicalutamide exist in the FDA Adverse Event Reporting System (FAERS) database. In all of the published case reports of liver toxicity with bicalutamide, the onset of symptoms was within the first 6 months of treatment. Symptoms that may indicate liver dysfunction include nausea, vomiting, abdominal pain, fatigue, anorexia, "flu-like" symptoms, dark urine, and jaundice. There are also published case reports of interstitial pneumonitis and eosinophilic lung disease associated with bicalutamide. along with hundreds of additional instances in the FAERS database as well. Interstitial pneumonitis can potentially progress to pulmonary fibrosis and may be fatal. Symptoms that may indicate lung dysfunction include dyspnea (difficult breathing or shortness of breath), cough, and pharyngitis (inflammation of the pharynx, resulting in sore throat). The exact incidence of liver toxicity and interstitial pneumonitis with bicalutamide are unknown, but both are said to be very rare events. A few cases of photosensitivity have been reported with bicalutamide. Hypersensitivity reactions (drug allergy) like angioedema and hives have also uncommonly been reported in association with bicalutamide. Because it is an antiandrogen, bicalutamide has a theoretical risk of birth defects like ambiguous genitalia in male fetuses. Due to its teratogenic capacity, contraception should be used in women taking bicalutamide who are fertile and sexually active.
=== 1 December === The RSF captured the 22nd Infantry Division headquarters in Babanusa and claimed full control over the city. The SPLM-N (al-Hilu) claimed to have retaken the town of Qardoud Nama in South Kordofan from the SAF.
== History of profession == The idea of physician extenders was conceived in 1966 by physician-educator Eugene A. Stead at Duke University, where the first physician assistant program was established. Three years later, also at Duke, Chairman of Pathology, Dr. Thomas Kinney established the first pathologists’ assistant program. As of June 2025, seventeen accredited programs have been established across the United States and Canada and six others are in various stages of accreditation.
In contrast to LAPs, where cyclodehydration occurs via the action of two distinct proteins responsible for leader peptide binding and cyclodehydrative catalysis, these are fused into a single protein (G protein) in cyanobactin and thiopeptide biosynthesis. However, in thiopeptides, an additional protein, designated the Ocin-ThiF-like protein (F protein) is necessary for leader peptide recognition and potentially recruiting other biosynthetic enzymes.

In CYP2D6 extensive metabolizers, the half-lives of 4-hydroxyatomoxetine and N-desmethylatomoxetine are similar at around 6 to 8 hours, whereas in CYP2D6 poor metabolizers, the half-life of N-desmethylatomoxetine is much longer at around 33 to 40 hours. Atomoxetine levels in cerebrospinal fluid (CSF) with atomoxetine at a dosage of 80 mg/day were 6.6 ng/mL at 8 hours post-dose and 1.4 ng/mL at 24 hours post-dose following 2 weeks of administration.
== January 15, 1982 (Friday) == The U.S. Federal Trade Commission dropped its 10-year-old antitrust lawsuit against the three largest makers of cereal in the nation— Kellogg, General Mills and General Foods. Hekuran Isai became the Minister of Internal Affairs for the People's Republic of Albania, as well as the director of the Communist nation's secret police, the Sigurimi, after the previous Internal Affairs Minister, Feçor Shehu, was arrested as part of a purge of government officials. The appointment came as the Albanian People's Assembly unanimously approved the appointment of Adil Çarçani as the new Prime Minister of Albania following the December 17 death by gunshot wound of Premier Mehmet Shehu. The Guatemalan Army invaded the village of Ilom (village) near Chajul and killed 28 men from the Mayan minority. A deadlier mass execution of 96 Mayan peasants would take place on March 23, 1982. The Lima Liturgy, drafted by Max Thurian, was celebrated in a multidenominational Christian gathering by the World Council of Churches Forbidden Broadway, an off-Broadway musical revue, written by Gerard Alessandrini and featuring parodies of current Broadway musicals, was given its first of 2,332 performances, opening at Palsson's Supper Club (now the Triad Theatre) in New York City. David Gibbons resigned as Premier of Bermuda and was succeeded by John Swan.
With the arrival of Mia and Chloe, producers took the opportunity to explore Ari's fictional backstory further, which Kipa-Williams relished. He thought Ari and Mia were "star-crossed lovers" and believed that Ari had never stopped loving her. The pair soon reconcile, but their relationship is tested by his involvement with a criminal gang and a targeted hit-and-run, which leaves him with serious injuries. Writers scripted a pregnancy for the couple, however, Mia is involved in a car accident during the 2021 mid-season finale, and she has a miscarriage. At the end of the year, Chloe's biological father Matthew Montgomery (James Sweeny) was introduced and Ari takes the blame for his death when Chloe accidentally kills him while protecting Mia. Kipa-Williams quit Home and Away and producers decided to kill-off his character. Ari is admitted to hospital with cancer of the appendix and he opts not to have surgery to remove the tumours, as he wants to die on his terms. Ari and Mia marry shortly before he dies. He made his final appearance on 15 March 2022. Kipa-Williams received a nomination for Best Daytime Star at the Inside Soap Awards in 2020. Ari and the Parata family received a positive response from critics and viewers, with some branding them New Zealand's answer to the River Boys, a popular group of characters introduced in 2011. The character's break-up with Mac was not well received, with Digital Spy's Susannah Alexander and Daniel Kilkelly saying the show had "lost a solid couple".
=== Neuronal membrane === The brain preferentially uses choline to synthesize acetylcholine. This limits the amount of choline available to synthesize phosphatidylcholine. When the availability of choline is low or the need for acetylcholine increases, phospholipids containing choline can be catabolized from neuronal membranes. These phospholipids include sphingomyelin and phosphatidylcholine. Supplementation with citicoline can increase the amount of choline available for acetylcholine synthesis and aid in rebuilding membrane phospholipid stores after depletion. Citicoline decreases phospholipase stimulation. This can lower levels of hydroxyl radicals produced after an ischemia and prevent cardiolipin from being catabolized by phospholipase A2. It can also work to restore cardiolipin levels in the inner mitochondrial membrane.

Primary drying removes ice by sublimation at low pressure and low shelf temperature. Secondary drying removes bound water by raising the shelf temperature, often under the same vacuum. The two stages differ in the water state being removed.
Freezing determines ice crystal size, pore structure, and the concentration of solutes in remaining liquid. Faster freezing generally creates smaller ice crystals and a denser dried matrix. These features affect drying rate and reconstitution behavior.
Lyophilization reduces water content but usually leaves a small amount of water in the dried material. Some water remains bound to solids or trapped in the dried matrix. Very low water targets can require extended secondary drying, which may alter product stability.
Most lyophilized products are stored in sealed containers at controlled temperature and humidity. Some require refrigeration, while others are stable at room temperature. Protection from light and moisture is often necessary.
